چکیده

Individual cone snail ( Conus sp.) contains thousands of bioactive peptides, but there are limited studies on its antimicrobial peptides (AMPs). Here, we investigated AMPs along with AMP-derived genes in the representative Chinese tubular C. betulinus ) by integration our previously published multi-omics (genomics, transcriptomics, and peptidomics) data. We identified a total 466 putative from genome, most them were annotated as histones . While at mRNA level, only 77 confirmed transcriptomic evidence, among Ubiquicidin, cgUbiquitin, Ap, VK10 abundant transcripts venom gland. In addition, 30 further validated peptidomics After silico analysis including 3D modeling, 11 chemically synthesized for an vitro assessment, eight proved good antifungal activity. summary, this work systematically characterized AMP repertoire This valuable genetic resource provides solid foundation in-depth therapeutic applications drug development.

High-throughput identification of novel conotoxins from the Chinese tubular cone snail (Conus betulinus) by multi-transcriptome sequencing.

BACKGROUND The venom of predatory marine cone snails mainly contains a diverse array of unique bioactive peptides commonly referred to as conopeptides or conotoxins. These peptides have proven to be valuable pharmacological probes and potential drugs because of their high specificity and affinity to important ion channels, receptors and transporters of the nervous system. Most previous studies ...

A Transcriptomic Survey of Ion Channel-Based Conotoxins in the Chinese Tubular Cone Snail (Conus betulinus)

Conotoxins in the venom of cone snails (Conus spp.) are a mixture of active peptides that work as blockers, agonists, antagonists, or inactivators of various ion channels. Recently we reported a high-throughput method to identify 215 conotoxin transcripts from the Chinese tubular cone snail, C. betulinus. Here, based on the previous datasets of four transcriptomes from three venom ducts and one...

High-Throughput Identification of Antimicrobial Peptides from Amphibious Mudskippers

Widespread existence of antimicrobial peptides (AMPs) has been reported in various animals with comprehensive biological activities, which is consistent with the important roles of AMPs as the first line of host defense system. However, no big-data-based analysis on AMPs from any fish species is available. In this study, we identified 507 AMP transcripts on the basis of our previously reported ...

High Throughput Identification of Antimicrobial Peptides from Fish Gastrointestinal Microbiota

Antimicrobial peptides (AMPs) are a group of small peptides, which are secreted by almost all creatures in nature. They have been explored in therapeutic and agricultural aspects as they are toxic to many bacteria. A considerable amount of work has been conducted in analyzing 16S and metagenomics of the gastrointestinal (GI) microbiome of grass carp (Ctenopharyngodon idellus). However, these da...

The Mitochondrial Genome of the Venomous Cone Snail Conus consors

Cone snails are venomous predatory marine neogastropods that belong to the species-rich superfamily of the Conoidea. So far, the mitochondrial genomes of two cone snail species (Conus textile and Conus borgesi) have been described, and these feed on snails and worms, respectively. Here, we report the mitochondrial genome sequence of the fish-hunting cone snail Conus consors and describe a novel...
